And will it be hard to get one then?

just be up and online ready for tix to go on sale and start trying maybe 10-15 mins before release time as both time i got tix i got them before 9:00am the day they were released

The email Glastonbury sent out last night says....

TICKET UPDATE

Following the ticket sale on October 4th, tickets for the 40th Anniversary Festival in 2010 are sold out. There will, however, be a re-sale of all cancelled orders early in 2010 (we'll announce the exact date and details nearer the time).

As that says "cancelled orders" I think it's pretty safe to assume that there won't be a sale until after the deposit tickets settlement date - which means early March.

Bizarrely this year I was through in 10 minutes! The previous three it's taken me normally 3 hours!

I reckon before the U2 announcement it would have been hard to get a ticket but not impossible, but now there will be a lot of U2 fans - never interested in Glastonbury otherwise - who will be looking to get tickets. Now it will just be very hard, but again, not impossible
